Western Blot HR substrate series products are chemiluminescent substances used to detect HRP-labeled antibodies during Western blotting. All such substrates consist of two parts, peroxide reagent and luminol/enhancer. Mix equal volumes of the two solutions, incubate with the blotted membrane, and then use the film or imaging system to detect the results.
Chemiluminescence measurement depends on the emission of light as a product of a chemical reaction, and is usually used for protein detection in Western blotting. The secondary antibody used in the Western blot is conjugated with horseradish peroxidase (HRP) enzyme, which reacts with the HRP substrate luminol. The reaction emits light at 428 nm, and the light signal can be captured on X-ray film or a digital imager.
However, luminol emits only weakly, so an enhancer is added to the reaction to increase the signal. This enhancement of the luminol-based signal is commonly referred to as enhanced chemiluminescence (ECL).

Abbkine SuperLumia ECL Plus HRP Substrate Kit is a value-priced, entry-level peroxidase substrate for enhanced chemiluminescence (ECL) that directly replaces costlier products without the need to re-optimize conditions.
- Mix the two substrate components at a 1:1 ratio to prepare the substrate working solution.
- Suggest 0.1-0.2ml of SuperLumia ECL Plus working solution for 1 cm2 size of blot membrane.
- Incubate blot 1-5 minutes in SuperLumia ECL Plus Substrate working solution.
- Drain excess reagent. Cover blot with clear plastic wrap.
- Expose blot to X-ray film.
Stable for one year at 4-8°C from date of shipment. Exposure to the sun or any other intense light can harm the working solution. Short-term exposure to typical laboratory lighting will not harm the working solution.
