Protein Purification
For analyzing individual proteins and protein complexes, identifying the interactions between proteins, proteins and nucleic acids, protein purification is the cornerstone of these studies. Since the purification of natural proteins is a challenging task, scientists have developed an experimental system that uses tag fusion proteins to capture, purify and detect the target protein. The most effective way to purify a protein is affinity chromatography, which achieves the purpose of purification by specific binding of the target protein to a matched immobilized ligand. The most notable feature is that the combination of a well-characterized tag and its corresponding solid-phase coupled ligand can achieve affinity purification of the labeled protein in a single step. Antibodies with fusion tags are also widely used in downstream detection and experiments, eliminating the need to develop corresponding probes for each specific recombinant protein.
Antibody purification mainly involves the selective enrichment or specific separation of antibodies from serum (polyclonal antibodies), ascites, and hybridoma cell line culture supernatants (monoclonal antibodies). Protein A, Protein G and Protein L are three bacterial proteins, which are well known for their efficient binding to antibodies. These proteins are recombined and expressed through genetic engineering editing, and are usually used for affinity purification of key antibody types from a variety of different species.
