Background Aspartate aminotransferase (AST, formerly known as glutamic-oxaloacetic transaminase, GOT, EC 2.6.1.1) is widely distributed in animal and plant tissues, predominantly in the liver, myocardium, skeletal muscle and kidney. Upon injury and rupture of tissue cells, large quantities of the enzyme are released into body fluids. Therefore, AST activity serves as an important biochemical indicator for evaluating the degree of tissue-cell damage. CheKine™Aspartate Aminotransferase (AST/GOT) Activity Assay Kit (IFCC Rate Method) can be used to detect biological samples such as animal and plant tissues, cells or bacteria, plasma, serum or other liquid samples. AST in the sample catalyzes the transamination reaction between L-aspartate and α-ketoglutarate to produce oxaloacetate and L-glutamate. Malate dehydrogenase (MDH) reduces the generated oxaloacetate to L-malate, accompanied by the consumption of NADH and formation of NAD+. NADH exhibits strong characteristic ultraviolet absorption at 340 nm, whereas NAD+ shows negligible absorption at this wavelength. AST activity can be calculated by measuring the decreasing rate of absorbance at 340 nm.
