Xanthine oxidase (XO, EC 1.17.3.2) is the terminal enzyme of purine catabolism in humans, catalyzing the hydroxylation of hypoxanthine to xanthine and then to uric acid[reference:76]. While XO is present in appreciable amounts in the liver and jejunum of healthy individuals, circulating XO levels increase dramatically in various liver disorders[reference:77].

How It Works

XO oxidizes xanthine to generate superoxide (O₂⁻)[reference:83]. O₂⁻ reacts with WST-8 dye to form a water-soluble colored formazan product, quantified at OD 450 nm[reference:84]. XO activity is proportional to the signal obtained[reference:85].
