Background

The mitochondrial respiratory chain is mainly composed of mitochondrial respiratory chain enzymes. Mitochondrial respiratory chain complex I (NADH oxidase) and mitochondrial respiratory chain complex II (succinate dehydrogenase) are the main elements for electrons to enter the mitochondrial electron transport chain (ETC). Complex I catalyzes the oxidation of NADH, and complex II catalyzes the oxidation of succinic acid to fumaric acid. Subsequently, coenzyme Q (Q) forms coenzyme (QH2), which eventually leads to a decrease in the terminal electron acceptor O2. Mitochondrial respiratory chain complex III (cytochrome c oxidoreductase) is an essential protein for mitochondrial oxidative phosphorylation. Mitochondrial respiratory chain complex III is the gatekeeper of the mitochondrial respiratory chain and the third main source of reactive oxygen species. Mitochondrial respiratory chain complex IV (cytochrome c oxidase) is the terminal electron acceptor of the mitochondrial electron transport chain. Complex IV converts O2 into water through the oxidation of cytochrome c. This process is related to mitochondrial cell membrane ATP synthesis. Mitochondrial respiratory chain complex V and the above four complexes complete oxidative phosphorylation to generate ATP, which is called ATP synthase, also called F1F0-ATPase.

Respiratory chain composition

The respiratory chain for respiration is composed of five protein complexes with relatively independent functions, namely complex I-V. Strictly speaking, the respiratory chain only contains complexes I to IV, and ATP synthase does not belong to the respiratory chain, but from the perspective of a complete ATP synthesis chain, ATP synthase is generally also called complex V.

Respiratory chain function

The mitochondrial respiratory chain is mainly related to energy metabolism and oxidative phosphorylation. The functions of its five components (the former: complex I~IV, the latter complex V) are as follows:

The former-the electron transport chain, oxidizes NADH and FADH2 produced by glycolysis and tricarboxylic acid cycle, while pumping protons out of the cell membrane or mitochondrial inner membrane to generate a proton gradient;

The latter-ATP synthase, also known as complex V, uses the influx of protons caused by the proton gradient to synthesize ADP and phosphoric acid into ATP, thereby storing the energy generated by hydrogen carrier oxidation in the form of ATP.
