| Applications notes | EliKine™ Rat Interferon Gamma (IFN-γ) ELISA Kit employs a two-site sandwich ELISA to quantitate IFN-γ in samples. An antibody specific for IFN-γ has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any IFN-γ present is bound by the immobilized antibody. After removing any unbound substances, HRP-Conjugated Rat IFN-γ detection antibody is added to the wells. Following a wash to remove any unbound HRP reagent, a Chromogen solution is added to the wells and color develops in proportion to the amount of IFN-γ bound in the initial step. The color development is stopped and the intensity of the color is measured. |

| Usage notes | • Do not mix components from different kit lots or use reagents beyond the kit expiration date. • Allow all reagents to warm to room temperature for at least 30 minutes before opening. • Pre-rinse the pipet tip with reagent, use fresh pipet tips for each sample, standard and reagent to avoid contamination. • Unused wells must be kept desiccated at 4 °C in the sealed bag provided. • Mix Thoroughly is very important for the result. It is recommended using low frequency oscillator or slight hand shaking every 10 minutes. • It is recommended that all samples and standards be assayed in duplicate or triplicate. |

| Background | Produced by lymphocytes activated by specific antigens or mitogens. IFN-γ, in addition to having antiviral activity, has important immunoregulatory functions. It is a potent activator of macrophages, it has antiproliferative effects on transformed cells and it can potentiate the antiviral and antitumor effects of the type I interferons. EliKine™ Rat IFN-γ ELISA Kit employs a double antibody sandwich method to quantitate Rat IFN-γ in samples. An antibody specific for Rat IFN-γ has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any Rat IFN-γ present is bound by the immobilized antibody. After removing any unbound substances, a HRP-conjugated antibody specific for Rat IFN-γ is added to the wells, form an antibody-antigen-enzyme-labeled antibody complex. Following a wash to remove any unbound streptavidin-enzyme reagent, adding HRP Substrate (TMB), TMB turns blue under the catalysis of HRP, and turns yellow after adding stop solution. Measure the OD value with a microplate reader at 450 nm wavelength. The IFN-γ concentration is proportional to the OD450 nm value. |
