Applications notes | CheKine™ Micro Total Iron-Binding Capacity (TIBC ) Assay Kit provides a simple, sensitive, rapid TIBC detection method, suitable for detecting serum samples. The principle is that under alkaline conditions, serum transferrin can be bonded to Fe3+, and the remaining unbound Fe3+ can be reduced to Fe2+, the bonded Fe3+ could be released after acidification ,and then be reduced into Fe2+ . A purple compound could be produced by the reaction between Fe2+ and philo oxazine ,and this purple compound has a special wave at 562 nm. Respectively reading the OD562nm values A1 and A2 , and A1 is based on the unbound Fe3+ , A2 is based on the bounded Fe3+ . And the concentration of TIBC is proportionate to the difference between A2 and A1. |

Usage notes | • Do not mix the components between different batch numbers and manufacturers; otherwise, the results may be abnormal. • Avoid bubbles while mixing or redissolving components. • Change pipette tips frequently to avoid cross contamination between components. • Ensure that all components and equipment are at the proper temperature before starting the experiment. • If the absorbance values is too high (>1.5). Pay attention to multiply by the dilution factor when calculating the result. • Please mix the standard well before use, it is normal while there is fewer precipitates in the standard solution. • Please take protective measures when operating, because ReagentⅡ has certain toxicity. |
