Applications notes The kit provides a simple method for detecting Phosphoenolpyruvate Carboxylase (PEPC) (EC 4.1.1.31) activity in a variety of biological samples such as animal tissues, plant tissues, cell culture: adherent or suspension cells, serum, plasma, bacterial. In the assay, Phosphoenolpyruvate carboxylase (PEPC) catalyzes phosphoenolpyruvate and CO2 to produce oxaloacetate and HPO42-, and malate dehydrogenase further catalyzes oxaloacetate and NADH to produce malate and NAD+. The rate of NADH decrease at 340 nm can reflect Phosphoenolpyruvate carboxylase (PEPC) activity.

Usage notes • Do not mix or substitute reagents or materials from other kit lots or vendors. Kits are QC tested as a set of components and performance cannot be guaranteed if utilized separately or substituted.
•Avoid foaming or bubbles when mixing or reconstituting components.
• To avoid cross-contamination, change pipette tips between additions of each standard level, between sample additions, and between reagent additions. Also, use separate reservoirs for each reagent.
•Ensure all reagents and solutions are at the appropriate temperature before starting the assay.
Storage instructions Storage at –20°C and Keep from light immediately upon receipt. Kit has a storage time of 6 months from receipt. Refer to list of materials supplied for storage conditions of individual components.

Additional Information

Background Phosphoenolpyruvate Carboxylase (PEPC) (EC 4.1.1.31) is widely present in animals, plants, microorganisms and cells. It is an enzyme that catalyzes the reaction of phosphoenol acetic acid with gas phase to produce oxaloacetic acid and presents an irreversible reaction. The operation of the acid cycle plays an important regulatory role.
