Applications notes CheKine™ Micro Advanced Oxidation Protein Products (AOPP) Assay Kit provides a simple, convenient, rapid and colorimetric total carbohydrate detection method. The principle of kit is the sample or standard reacts with the initiator and stop solution, and the product has an absorption peak at 340 nm. The content of AOPP in the sample is determined by detecting the OD value at 340 nm and comparing it with the standard curve.

Usage notes • Do not mix the components between different batch numbers and manufacturers, otherwise, the results may be abnormal.
• Avoid bubbles while mixing or redissolving components.
• Change pipette tips frequently to avoid cross contamination between components.
• Ensure that all components and equipment are at the proper temperature before starting the experiment.
• In order to guarantee the accuracy of experimental results, need to do a pre-experiment with 1-2 samples.
• If the OD value exceeds 1.3, the sample needs to be diluted.
• The sample is hemolyzed or has a dark color and needs to be compared with itself. The specific method is to add 200 µL of the diluted sample and 30 µL of Extraction Buffer (1x) to UV microplate, incubate at 37°C for 5 min, record the absorbance at 340 nm, and subtract the OD value of the self-control from the measurement well during calculation.
• Serum, plasma and tissue samples are recommended to be diluted 5 times with Extraction Buffer (1x) for testing.
• The Reagent I and Reagent II are stored in sealed condition.
Storage instructions Storage at –20°C and Keep from light immediately upon receipt. Kit has a storage time of 6 months from receipt. Please refer to the table below Materials supplied and Storage conditions to store all the components.

Additional Information

Background Advanced Oxidized Protein Products (AOPP) is a newly discovered class of uremic toxins with pro-inflammatory activity, and is an oxidative modification product formed after plasma proteins are attacked by reactive oxygen species (ROS). AOPP can stimulate monocytes and neutrophils, stimulate the synthesis and release of a large number of pro-inflammatory cytokines, mainly tumor necrosis factor TNF-ɑ, and promote the production of more reactive oxygen species, thereby inducing and aggravating systemic oxidative stress And inflammation. AOPP is very similar to Advanced Glycation End-Product (AGE) proteins, and they also perform similar biological functions. AOPP is an important detection index for diseases such as renal complications, atherosclerosis, diabetes, metabolic syndrome, immune diseases and malignant tumors.
