Product name Rat Mitochondrial import receptor subunit TOM34 (TOMM34) ELISA Kit
Reactivity Rat
Applications ELISA
Applications notes This Rat Mitochondrial import receptor subunit TOM34 (TOMM34) ELISA Kit employs a two-site sandwich ELISA to quantitate TOMM34 in samples. An antibody specific for TOMM34 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyTOMM34 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for TOMM34 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of TOMM34 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Detection method Colorimetric
SampleType Cell culture supernatants, Other biological fluids, Plasma, Serum
Assay type Sandwich ELISA (quantitative)
Assay duration Multiple steps standard sandwich ELISA assay with a working time of 3-5 hours. It depends on the experience of the operation person.
Alternative TOMM34; HTOM34P; TOM34; URCC3; mitochondrial import receptor subunit TOM34; outer mitochondrial membrane translocase (34kD)

Additional Information

Background TOMM40 is the channel-forming subunit of the translocase of the mitochondrial outer membrane (TOM) complex that is essential for protein import into mitochondria. The deduced protein contains 361 amino acids. Fractionation of rat liver detected Tomm40 within the mitochondrial fraction. Extraction and antibody-accessibility studies suggested that Tomm40 is embedded within the mitochondrial membrane.Tomm40 was associated with a 400-kD complex containing the mitochondrial import receptors Tom20 and Tom22 and several smaller components. Mitochondria-targeted preproteins interacted with the Tomm40-containing complex during their transit to the mitochondrial matrix, and immunodepletion of Tomm40 caused the loss of preprotein import activity.
