Applications notes | CheKine™ Micro Ribulose bisphosphate carboxylase/oxygenase (Rubisco) Assay Kit provides a simple method for detecting Rubisco activity in plant tissues. One molecule of ribulose-1,5-diphosphate (RuBP) combines with one molecule of CO2 to produce two molecules of 3-phosphoglyceric acid (PGA). Glyceraldehyde-3-phosphate can be produced by PGA through the action of additional 3-phosphoglycerate kinase and glyceraldehyde-3-phosphate dehydrogenase, which is accompanied by NADH oxidation to produce NAD+. NADH has a characteristic absorption peak at 340 nm, while NAD+ does not. Rubisco activity is calculated by measuring the rate of decrease in light absorption at 340 nm. |
