Additional Information

Background Copper is an essential trace transition metal in eukaryotic cells, acting as a cofactor for key enzymes including cytochrome c oxidase and SOD1. Its intracellular homeostasis is tightly regulated and critical for mitochondrial respiration, antioxidant defense, neurotransmitter synthesis, and iron metabolism. Both copper deficiency and excess impair cellular function by inducing oxidative stress and disrupting mitochondrial metabolism. Additionally, copper participates in hematopoiesis by catalyzing hemoglobin synthesis, and supports skeletal and neural development. Thus, accurate quantification of total copper is fundamental to understanding copper homeostasis. This kit employs a specific colorimetric assay under acidic conditions, copper ions are fully released from copper-binding proteins (e.g., ceruloplasmin), and in a reducing environment, they form a stable colored complex with the highly selective chelator 3,5-DiBr-PAESA, with maximal absorbance at 575–585 nm. The absorbance correlates linearly with total copper concentration over a defined range. Quantification is achieved by measuring absorbance on a microplate reader against a standard curve.
