Additional Information

Background The cytochrome P450 enzyme superfamily is a class of heme-containing monooxygenases widely present in living organisms, playing critical roles in drug metabolism, xenobiotic detoxification, and endogenous substance synthesis. Their most prominent characteristics include broad substrate specificity, diverse catalytic reactions (e.g., hydroxylation, dealkylation, epoxidation, etc.), and an electron transport chain dependent on reduced nicotinamide adenine dinucleotide phosphate (NADPH) and molecular oxygen. Different isoforms of P450 enzymes exhibit distinct tissue distributions and substrate selectivities, and are often used as biomarkers for assessing drug–drug interactions and the toxicity of environmental pollutants. The principle is: Cytochrome P450 enzymes possess 7-ethoxycoumarin O-deethylase (ECOD) activity, catalyzing the O-deethylation of the substrate 7-ethoxycoumarin to produce the highly fluorescent metabolite 7-hydroxycoumarin. The catalytic activity of CYP450 enzymes can be quantitatively determined by measuring the increase in fluorescence intensity of 7-hydroxycoumarin in the reaction system using a fluorometer.
