Background Total bile acid (TBA) is mainly used for screening and prognostic follow-up of hepatobiliary diseases and used as a marker of liver parenchymal damage and cholestasis. Viral hepatitis, cirrhosis, alcoholic liver disease, drug-induced liver injury, cholestasis, etc. often lead to an increase in serum total bile acid levels. The CheKine™ Total Bile Acid (TBA) Content Assay Kit can detect the total bile acid content in serum or plasma. The principle is that the bile acid is specifically oxidized by 3 α -hydroxysteroid dehydrogenase (3α -HSD) and oxidized β -thionicotinamide adenine dinucleotide (Thio-NAD+ ) to generate 3-ketosteroids and reduced β -thionicotinamide adenine dinucleotide (Thio-NADH). The 3-ketosteroids react with 3 α -HSD and reduced β -nicotinamide adenine dinucleotide (NADH) , and generate bile acids and oxidized β-nicotinamide adenine dinucleotide (NAD+ ) . Within a certain period of time, the Thio-NADH produced by the enzyme cycle is directly proportional to the bile acid content in the sample. By detecting the changes in Thio-NADH absorbance at a wavelength of 405nm, the total bile acid content in the sample can be calculated.
