The principle is that amylase catalyzes the hydrolysis of starch to produce reducing sugar, which reduces 3, 5-dinitrosalicylic acid to produce brown-red material, with an absorption peak at 540 nm. Amylase activity was calculated by measuring the absorbance increase rate at 540 nm. |

| Background | Amylases are responsible for the hydrolysis of starch, mainly consisting of α -amylase (α -Al) and β -amylase (β -Al, EC 3.2.1.2). β -amylase cleaved the α-1, 4-glycosidic bond from the non-reducing end of starch to produce reducing sugars such as glucose, maltose, maltose trisose and dextrin. |
