| Background | Apoptotic cells undergo a number of important changes, including cell shrinkage, cytoplasmic agglutination, DNA disruption, and formation of membrane vesicles, leading to the separation of cell components to form apoptotic bodies, which are then phagocytic by macrophages or parenchyma cells. The Apoptosis Detection Kit detects apoptosis based on changes in phosphatidylserine (PS) turnover of cell membrane and nuclear rupture. |

Before preparing the TdT labeling reaction buffer, equilibrate each component to room temperature. The Equilibration Buffer (5×) stock solution may be separated due to low temperature storage. Please invert and mix well before use.
Both TUNEL and Annexin experiments can used A and B inducer. Its purpose is to induce apoptosis, which can be used alone or in combination. Both A and B can induce apoptosis in some common cells. A and B complement each other and can induce apoptosis in more types of cells.
Cell fixation is required, and antigen retrieval is not required. This product marks the DNA fragmented in the nucleus caused by apoptosis, rather than antigenic proteins.
It is recommended to wash at least 2-3 times, because there are many impurities in the medium, which will stick to the cell membrane. After washing, centrifuge well and remove the supernatant with care.
Wash at room temperature at an interval of 5 minutes at a time.
