Product name Human Tryptase beta-2 (TPSB2) ELISA Kit
Reactivity Human
Applications ELISA
Applications notes This Human Tryptase beta-2 (TPSB2) ELISA Kit employs a two-site sandwich ELISA to quantitate TPSB2 in samples. An antibody specific for TPSB2 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyTPSB2 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for TPSB2 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of TPSB2 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Detection method Colorimetric
SampleType Cell culture supernatants, Other biological fluids, Plasma, Serum
Assay type Sandwich ELISA (quantitative)
Assay duration Multiple steps standard sandwich ELISA assay with a working time of 3-5 hours. It depends on the experience of the operation person.
Alternative TPSB2; TPS2; tryptaseB; tryptaseC; OTTHUMP00000208944; mast cell tryptase beta; mast cell tryptase betaI; tryptase; tryptaseI; tryptase beta 2; tryptase beta; tryptase betaI

Additional Information

Background Tryptases comprise a family of trypsin-like serine proteases, the peptidase family S1. Tryptases are enzymatically active only as heparin-stabilized tetramers, and they are resistant to all known endogenous proteinase inhibitors. TPSb2 is characteristic of tryptases but is unusual in other genes. The alleles of this gene exhibit an unusual amount of sequence variation, such that the alleles were once thought to represent two separate genes, beta II and beta III. Beta tryptases appear to be the main isoenzymes expressed in mast cells, whereas in basophils, alpha-tryptases predominate. Tryptases have been implicated as mediators in the pathogenesis of asthma and other allergic and inflammatory disorders.
